1960年代、ラットを用いた動物実験により「膀胱がんのリスクが高くなる」という研究が発表され、多くの国での使用禁止となりました。しかし、その後の研究により、発がん性はサッカリンを合成する過程で生成された不純物オルトトルエンスルホンアミドによることが判明し、サッカリンに発がん性はないとの見方が優勢となりました。その結果、米国や中国ではサッカリンが飲食物に再び使われるようになりました。
